Cell density solution?

In my last post I talked about how figuring out if cell density was causing the delayed growth would be tricky. I cant just plate large vs small numbers because the large ones would just lawn. And we don’t know how to actually make cells comptent, at most a small fraction of them will be transformed. We have higher competent mutants like sxy-1 but it’s still not great

I just had a thought, might be a simple solution

Grow up Nov resistant wildtype and Nov sensitive wildtype cells. Put a small amount of Nov-r into the Nov-s culture, same proportion that is usually transformed. Vortex. Plate.  Now I can test to see if the delayed growth is being caused by the competent cells themselves rather than variables
